First the necessities,
Pre-paid sim:
If you fly Singapore Airline, you can get a tourist repaid card on board so that when you land, you are ready to go.
Alternatively, you can get your sim from any 7-eleven stores. Remember to bring along your identification card/Passport as that is required by law.
Where to stay
Ok, this is very subjective and of course depends on your budget. My tip is would be as a traveller, anywhere near a MRT is all you need unless you are happy to taxi around then it doesn't really matter where you stay. Most of the places of interest are located around the city center such as marina, orchard and sentosa.
Note: Staying in sentosa can be troublesome for visiting the mainland because of transport. You will have to catch a train to vivocity and transfer over to the mrt on mainland. If you intend to spend alot of time on sentosa such as visiting the casino and all the theme park, it may be worthwhile considering booking a hotel for a couple of nights here. For just the theme park and the aquarium, a day trip is more than enough to cover.

This time round I am planning to visit
Singapore Zoo
One of the best zoo in the world and they have awards to prove it. The zoo has been designed to mimic the animal's natural surrounding as closely as possible and it covers over 300 different species of animal.

I have been there before but would definitely love to revisit again as I did recall having a pretty good time there.
River Safari
Next would be the River Safari. I have not been so I cannot comment however this can be purchased as part of a package granting access to a total of four park for $69 which includes the singapore zoo, jurong bird park, river safari and night safari.

Night Safari
Again, I have not been but as far as I know, this is similar to the zoo except you will have the opportunity to see more nocturnal animals active.
As the world's first safari park for nocturnal animals, this is where you can explore the rainforest at dusk, enjoy tribal performances, and more.
Gardens by the bay
Wikipedia says it all
Gardens by the Bay is a nature park spanning 101 hectares of reclaimed land in central Singapore, adjacent to the Marina Reservoir. The park consists of three waterfront gardens: Bay South Garden, Bay East Garden and Bay Central Garden
Malaysia: Johor bahru
Yes I know, it is not Singapore however it is just across the border. We go in mainly to eat, massage and shop because it is cheaper than Singapore. Durian anyone?

National Museum
Entry fee is cheap and its the best way for visitor's to gain an insight to the colorful past of Singapore.
Universal Studio
If you have never been to any universal studio in the world, I highly recommend you visit this in Sentosa. Out of all the rides, the 2 that you CANNOT SKIP are
- The mummy returns
- Transformer ride
Chinatown/Buddha Tooth Relic Temple
Spend an hour or 2 here just to absorb some of the cultural, food and tradition atmosphere. Do however note everything here is jacked up to maximise the price gorging on tourist. The striking difference between chinatown juxtapose against the modern skyline makes a great picture if you can picture it at the right time, place and angle.
Do not forget to visit the Buddha Tooth Relic temple whilst you are there!Southern Ridges Hike/Trail
A pretty nicely done walking trail
This 10-km stretch of connecting trails lets you walk among the trees to observe a wide variety of flora and fauna in their natural habitat.
Singapore Flyer/Marina bay
The only reason why I would recommend this place would be for some photo taking session. this place is easily accessible however, I wouldn't plan alot of time here. A trip to singapore wouldn't be complete with the iconic snap of the merlion against the backdrop of the Flyer and Sands casino.
SEA Aquarium
lastly, the SEA Aquarium which is also on Sentosa right beside universal studio. Make sure you plan these 2 together. The aquarium can comfortably be covered in 2 hours. This is at one time the biggest aquarium in the world by volume and species but has since been taken over by China :(

Aquarium (South East Asia Aquarium) was the world's largest aquarium by total water volume until overtaken by Chimelong Ocean Kingdom in Hengqin, China.[4] It contains a total of 45,000,000 litres (9,900,000 imp gal; 12,000,000 US gal) of water for more than 100,000 marine animals of over 800 species.[1] The aquarium comprises 10 zones with 49 habitats. The centerpiece of the Aquarium is the Open Ocean tank with more than 18,000,000 l (4,000,000 imp gal; 4,800,000 US gal) and 50,000 animals.[5] Until 2014 when eclipsed by China's Chimelong Ocean Kingdom, it had the world's largest viewing panel, 36-metre (118 ft) wide and 8.3-metre (27 ft) tall, which is intended to give visitors the feeling of being on the ocean floor.[2]
